I have a few offers that expire on April 1. If I accept one on OLSAS, would that automatically take me out of the running at other schools & prevent any other offers from coming in?

As per OLSAS website:

"Note: When you provide a firm acceptance, your other choices will be marked as “No Further Consideration”. Your application to any other Ontario law school(s) will be cancelled, and you will be removed from all wait lists."

you can have a provisional acceptance which will still allow you to be considered by other schools.
